苦茶Camellia assamica var.kuchaChang et Wang的嘌呤生物碱 (1)

摘要:用重量法和柱层析法从苦茶Camellia assamica var.kucha Chang et Wang的嫩梢中分离出3种嘌呤生物碱,经薄层层析、熔点测定、元素分析、核磁共振和质谱等对其结构的测定,鉴定为咖啡碱、1,3,7,9-四甲基尿酸和可可碱,其百分含量分别为0.88%,0.84%和0.29%.样品水提物经氯仿萃取后直接由高压液相色谱分析,测得苦茶含咖啡碱1.932 9%,四甲基尿酸1.294 1%,可可碱0.585 5%,茶叶碱0.012 8%.这是茶组植物中嘌呤生物碱新的分布模式.

Purine Alkaloids in Camellia assamica var.kucha Chang et Wang

Abstract:Purine alkaloids in Camellia assamica var.Kucha Chang et Wang were investigated by extraction combined with silica gel column chromatography. Three products were isolated. Product A and C were determined as caffeine and theobromine by TLC and melting point compared with standards. The structure of product B were determined by MS and 1HNMR, the element analysis and melting point for 1,3,7,9-tetramethyluric acid.The purine alkaoids were consisted of caffeine,1,3,7,9-tetramethyluric acid, theobromine and theophylline in the leaves of C.assamica var.kucha, and their content was 1.932 9%, 1.294 1%, 0.585 5% and 0.012 8% respectively by HPLC.The distribution of new type of purine alkaloids and such high content of tetramethyluric acid in wild tea tree resources is firstly reported.

山茶属Camellia约280种,其中茶组Camellia Sect.Thea L.有32种,著名的饮料茶叶就是来自茶组植物的茶C.sinensis(L.)O.Ktze和普洱茶C.assamica(Mast.)Chang.茶叶含有复杂的化学成分,其中嘌呤生物碱是茶最重要的化学成份之一,其他的化学成份尚有茶氨酸、儿茶素、芳香油、色素、糖类、有机酸、维生素类、酶类、水浸出物和矿质元素等.在茶叶商品流通中,把咖啡碱、锰、儿茶素和茶氨酸列为辨别真假茶必检的项目.可见咖啡碱在茶叶中生化成份检测中的地位[1].茶叶的生物碱主要为嘌呤碱,也有少量的嘧啶碱.在茶叶中发现的嘌呤碱有咖啡碱、可可碱、茶叶碱、腺嘌呤、鸟便嘌呤、黄嘌呤、次黄嘌呤和拟黄嘌呤等.其结构特点系以嘌呤环为基本骨架,不同种类的嘌呤碱取决于嘌呤环上甲基的位置和个数.茶叶生物碱中,咖啡碱占干质量2%~4%,可可碱约0.05%,茶叶碱约0.002%.目前在茶组植物中,已经发现了和茶不同的嘌呤碱分布模式,这就是以可可碱为主要成份的缅甸茶C.irrawadiensis Barua、紫果茶C.purpurea Chang et Chen和可可茶C.ptilophylla Ching[2]的芽叶含可可碱为主,其中可可茶含可可碱超过干质量的4%,最高达6.84%[3~7].最近在研究苦茶C.assamica var.kucha Chang et Wang时,分离出具有意义的较高含量的1,3,7,9-四甲基尿酸.下面是研究的初步结果.

1样品和试剂、仪器

1.1供试样品

采摘1芽2叶苦茶鲜样,经隔水蒸15 min,在80 ℃烘箱中烘干,置于放有硅胶的干燥器中备用.栽于中山大学实验园地的苦茶是1986年由采自原产地的种子实生苗成长的,树高2 m左右.
